Genetic relationships among Arachis species based on AFLP Genet. Mol. Biol.

Amplified Fragment Length Polymorphism (AFLP) was used to establish the genetic relationships among 20 species from seven of the nine sections of genus Arachis. The level of polymorphism among nine accessions of the cultivated peanut, A. hypogaea L., was also evaluated. Three combinations of primers were used to amplify the AFLPs. The fragments were separated in 6% denaturing acrylamide gels. A total of 408 fragments were analyzed. An average of 135.3 fragments per primer combination were scored, and the largest number of fragments was 169 using primer combination Eco RI - ACC / Mse I - CTG, while the lowest was 108, with Eco RI - ACT / Mse I - CTT. The amplified fragment length polymorphism (AFLP) assay is an efficient method for the identification of molecular markers, useful in the improvement of numerous crop species. Bulked Segregant Analysis (BSA) was used to identify AFLP markers associated with water-stress tolerance in barley, as this would permit rapid selection of water-stress tolerant genotypes in breeding programs. Objetivou-se, com este trabalho, verificar a variabilidade morfocultural e genética de fungos associados à podridão peduncular do mamão (Carica papaya L.). As avaliações morfoculturais foram feitas pelo índice de crescimento micelial e características das colônias, como coloração e tipo de micélio, quando os isolados foram crescidos em diferentes meios de cultura (BDA, MDA e Czapek). The objective of this study was to evaluate whether different similarity coefficients used with dominant markers can influence the results of cluster analysis, using eighteen inbred lines of maize from two different populations, BR-105 and BR-106. O objetivo do trabalho foi analisar a estrutura e a diversidade molecular de quatro populações de Spodoptera frugiperda (J.E. Smith) associadas às culturas de milho e arroz no Rio Grande do Sul. Foram coletadas lagartas de quatro populações em áreas isoladas (distanciadas em mais de 300 km) nos municípios de Santa Rosa (milho) e Uruguaiana (arroz irrigado) e em áreas adjacentes, no município de Pelotas (milho e arroz irrigado). The Amplified Fragment Length Polymorphism (AFLP) technique was used to access genetic diversity between three domestic and nine wild proso millet biotypes from the United States and Canada. Eight primer combinations detected 39 polymorphic DNA fragments, with the genetic distance estimates among biotypes ranging from 0.02 to 0.04. Colorado-Weld County black seeded and Wyoming-Platte County were the most distinct biotypes according to the dissimilarity level. Somaclonal variation in plants regenerated by organogenesis from long-term cultured calluses of two diploid staminate genotypes of Asparagus officinalis cv. Argenteuil was characterized by plant phenotype, ploidy, meiotic behavior, pollen viability, fruit and seed set, and AFLP profiles. Phenotypic deviations from the donors were detected in foliage color, flower size, and cladode and flower morphology. Ploidy changes were observed in 37.8% of the 37 regenerants studied. Identificación de Cultivares y Líneas de Mejoramiento de Arroz de Chile Mediante Amplificación de Fragmentos Polimórficos (AFLP) Agricultura Técnica

Doce cultivares y líneas de arroz (Oryza sativa L.) desarrollados por el programa de fitomejoramiento del Instituto de Investigaciones Agropecuarias (INIA), Centro Regional de Investigación Quilamapu, fueron caracterizados genéticamente mediante amplificación de fragmentos polimórficos (AFLP). De 21 combinaciones ensayadas, sólo 16 fueron informativas, generando un total de 667 amplicones, 94 de ellos polimórficos (14,4%), con un rango entre 9 y 33% de polimorfismo por combinación. In this study, a total of 14 agronomic traits, five AFLP primer combinations and ten SSR loci were used to describe and to classify a group of Tunisian olive genotypes into groups based on molecular profiles and agronomic traits. The analysis of variance of the agronomical data revealed significant differences among accessions for all measured traits. The mean phenotypic dissimilarity (0.34 with a range of 0.08-0.6) was low in comparison to dissimilarity calculated using AFLP (0.50 with a range of 0.16-0.70) and SSR markers (0.76 with a range 0.35-0.94). Genetic similarity among soybean genotypes was studied by applying the amplified fragment length polymorphism (AFLP) technique to 317 soybean cultivars released in Brazil from 1962 to 1998. Genetic similarity (GS) coefficients were estimated using the coefficient of Nei and Li (Nei and Li 1979), and the cultivars were clustered using the unweighted pair-group method with averages (UPGMA). The parentage coefficients of 100 cultivars released between 1984 and 1998 were calculated and correlated with the genetic similarity obtained by the markers. Use of single-enzyme amplified fragment length polymorphism for typing Clostridium perfringens isolated from diarrheic piglets BJM

Clostridium perfringens is an important pathogen in human and veterinary medicine. In swine, the agent is responsible for necrotic enteritis and enterotoxemia characterized by diarrhea, weight loss, delayed development and, in some cases, death. In the present study amplified fragment length polymorphism analyses (AFLP) was used to characterize 54 C. perfringens strains isolated from swine presenting diarrhea. Analysis of the results showed 29 distinct profiles with discriminatory index equal to 0.97. A first generation genetic linkage map of the European flat oyster Ostrea edulis (L.) based on AFLP and microsatellite markers ArchiMer

AFLP and 20 microsatellite markers were genotyped in a three-generation pedigree comprised of 2 grand-parents, 2 parents and 92 progeny. AFLP-based genetic linkage maps of the blue mussel (Mytilus edulis) ArchiMer

We report the construction of the first genetic linkage map in the blue mussel, Mytilus edulis. AFLP markers were used in 86 full-sib progeny from a controlled pair mating, applying a double pseudo-test cross strategy. Thirty-six primer pairs generated 2354 peaks, of which 791 (33.6%) were polymorphic in the mapping family. Among those, 341 segregated through the female parent, 296 through the male parent (type 1:1) and 154 through both parents (type 3:1).
